Repeated injection of 40μL of 0.6 mg/mL biotinylated albumin injected on Sepharose 4B Streptavidin beads at a flow rate of 1μL/sec. Assuming that absorbance of b-BSA is the same in solution and bound on the beads, 2.6μg of albumin was bound to bead surfaces during the first injection corresponding to 10% immobilization efficiency, while the second injection resulted in only 3% immobilization efficiency, while binding capacity has apparently been reached. This is, of course, only an estimate and the binding capacity at equilibrium obtained in batch mode would most likely be higher.
